Hi.

If I buy a drilled tank and don't want to use an overflow, is there a way I can reliably plug the holes?

you might be able to take a piece of glass a good bit bigger than the hole and silicone it to the inside of the tank over the hole... just be sure you have a seal all the way around and a good overlap to spread the push on the glass around

If the bulkhead fitting through the wall or floor of the tank is still there, screw cap blind plugs will be available to plug it. I would have a lot more confidence in that than in a glass patch. Your choice whether the the seal is removable or permanent.
