I need to install RO/DI system and don't know where the whole in the drain should be. if someone knows for sure, please point me in the right direction.

If you're drilling a hole to install a clamp on drain fitting, like this one, you want to install it as far below your RO/DI unit as possible to allow for gravity to work and so it can drain freely (back pressure on the drain line isn't really desireable). At the same time, since you're drilling a hole in the plumbing, it's generally a good idea to install it in a section of the pipe that can be easily replaced or a repair can easily be spliced in, just in case you decide to uninstall the RO/DI unit from that location.

reef has a good idea but im in plumbing repair and trust me you dont want to drill through a fixed drain line like fsn said drill through a piece that is easily replaced in case you mess up or it need to be removed i would go in the j of the s trap thats the piece right below the straight piece on the sink side
