Want to Change Substrate

I have a mixed CC and live sand substrate in my 20g and I wish I had only used sand. I recently set up a 5g refugium with 10lbs of seeded sand.

My 20g is established, but I want to change the substrate. I would like to allow the sand in the refugium to age for a month or so and then use it to seed a new sand substrate in my established 20g.

Does anybody think that my tank will suffer if I quickly remove the current substrate and add the sand from my refugium?

Depends on how you do it. I swapped CC for sand and had no problems, but I was putting in fresh sand, so there was no disturbance of bioldogical layers. In your case, I would put in fresh sand to the main tank and then just seed it with a cup or 2 from the fuge, rather than moving all of it from the fuge into the tank. That leaves a healthy amount of the biofiltration intact.
