Skim just the surface of the sand and, if needed, regulate the flow by the ball valve.

The main possible problem with sand is formation of anaerobic spots leading to production of poisonous hydrogen sulfide gas. To avoid this, keep your sand layers thin and mix the sand with your hand or vacuum tube regularly. For a river sand, I'd avoid layers thicker than about 1 1/2" even in planted tanks. Non-planted tanks don't need that much, of course. And the larger the sand particles are, the better.
