It may be time to bleach it. This will do two things, first it will make your python look better and second it should kill off most of the mildew. Bleaching does not rid your Python of the mildew, it merely kills it and makes it less noticeable.

Before proceeding please read and understand all warnings on the bleach bottle. Use the utmost care when handing this chemical.

This procedure is most easily done in a bath tub. First remove all the water from the hose and remove the BIG vac tube and adapter from the ends of the hose. Put one end of the hose near the drain in the tub or sink and leave it there. Use a funnel and pour enough bleach into the other looped end of the hose to fill a few inches of hose.

Now keep a loop drooping down and alternately raise and lower each side of the loop so as to coat the inside and bathe the hose with the bleach. You will notice the mildew change from black to mostly clear. As you clear up a length of hose move the bleach along to a new area to bleach. Keep going until the whole hose is done. Leave it in the tub or sink for a few minutes and then rinse by running water through the Python for a few minutes until the bleach smell is gone. You may want to run some dechlorinator through the hose to be sure no chlorine is left.

Great one! I sometimes just leave both ends up at the sink, the rest layed out on the floor. The use a pitcher to fill the hose with bleach water until it comes back up the other end.
 
Thanks for the tip! I feel silly now for throwing away my old water vac when it got mildewed....
