Might be calcium/hard water build up. You can try cleaning it with some vinegar and a razor blade.

You might want to look up columnaris. See if those pictures are what your gup had.

When you added the fish did you do daily testing? It may have gone into a mini cycle adding fish. So your water parameters may have been that prior to adding but werent after. You weren't clear when you did the testing. How did you acclimate the guppies?

Im slightly concerned why you have no nitrates if your tank is cycled and that's a false ammonia reading. Do you have a lot of real plants? It didnt look like it from the picture but obviously can't see the whole thing.

Go to your local pet store, look in the bird dept for shell grit. It's crushed shell that has already been cleaned, it's what i use in a media bag in my filters. About a handful for 10 gallons. It takes a few weeks for it to work though so be patient.

Prime doesnt deal with any heavy metals though. So if you use tap water/city water/well water that is something you want to keep in mind.
