Is it safe to use by itself? does it need a layer of gravel on top?
BTW I have a 40G long

IF you are talking about Schultz's Soil Conditioner, also known as Profile, yes it's absolutely safe. You can mix it with another substrate or Flourite(what I did), or use it by itself.
It's a bit lighter in color than Flourite and also lighter in weight due to it's higher porosity. It moves around a little bit when gravel siphoning, due to it's lightness but that, to me is no big deal. It's increased porosity gives it the ability to house the bacteria that all tanks need. It's very, very similar in composition to Flourite at a fraction of the cost.

Would a 2/2 mix with flourite be okay?

That should work well. You can layer it or mix it up.
