Switching from a penguin 330 to a aquaclear 500..do I just start using the new filter? or should I try to put my old penguin cartridge in the new aquaclear 500 to keep the bio filteration going? If I just start the new filter up will my tank go into a mini cycle?

Yes, removing the existing filter will initiate a cycle, quite possibly a large one, especially if this is an unplanted tank.

If the filter media are compatible, using the existing media in the AC, then gradually removing it and replacing it with AS media (sponges or noodles, etc) will work. Otherwise, run them in tandem for 2 weeks, then gradually remove the media from the old filter until it's all gone, and the AC media is supporting the full load.

Good advice. I like the idea of running both filters in the beginning. It is the easiest way if they both fit on the back of the tank. How big is the tank anyway and what fish are in it?

I agree with OrionGirl, run the 2 filters together for 2 weeks or so. I'm not sure if the penguin 330 media would fit your aquaclear 500... but being quite big, it just might. In that case, transfer all your penguin media (do not clean the cartridges!) to the AClear, and no fish will see the difference.
