Would a handful be enough to help jumpstart the biofilter?

Any amount will help jumpstart the cycle, the more you do the more effect it will have. if it comes from a tank with no UGF, the upper level of the gravel will be the most beneficial as the lower you go the bacteria is limited by oxygen.

It would certainly help. How much so, depends on what part the gravel has in the biological filtration. If the gravel is from a UGF set up then quite a bit more so than a non-UGF tank.

But, even if not, it will assist in the cycling of a new tank.

If you have another type filter, then a squeeze of the biological media from the established tank's filter would help quite a bit more.
 
It's even good to add a cupfull or two of gravel/sand from any other established tank, also. That tank might have a different spectrum of flora that might not be in your tank. You can't have too much bio-diversity.

Maybe a handful of filter noodles would help to a greater extent. I tend to do this for some of my friends seemed to work well and I always get a box of new noodles in return :p Just make sure you don't remove too much!
