Yes. This is done quite commonly, especially in planted tank where some people have reservations with running RUGFs in a aheavily planted tank.
For example, I have a 55G UGF Plate in my 90G tank. Works great and still allows me to have the benefits associate without.
HOWEVER, a word of caution here as well... The reaosn for setting up a full-tank plate is to prevent particulates, roots, etc. from getting under the plate itself and blocking the pathways. As a result I used some aquarium silicone and extra acrylic on my plate to block the exposed end. This with the fact that I am not drawing water as is typical in a UGF, but pushing water through the plate via my RUGF, I feel confident that this is okay.
I would not do this with a traditional UGF system, but I'm sure it can be done.
