Marineland advises to put a full but reasonable fishload in the tank at the same time as Bio-Spira. If those fishes are going into the 20g tank, then you can add all of them at once.
Just follow the directions on the package. Do not deviate from the instructions. Do not put the Bio Spira in your filter, put it in the tank like the instructions tell you.
The only thing I would add that are not in the instructions is to not do *any* water changes for at least 48 hours after you add the Bio Spira. It takes at least that long to colonize and you will just throw out the bacteria if you do.

The directions for use are on the bottom. Do not use Bio Coat. Your fish do not need to be irritated to produce slime coat. It's just a marketing speil. You can use either Prime or Bio Safe. Marineland will not give the "okay" to Amquel or any other product.
good to hear i don't have to buy bio-coat, less money to spend! i guess i will be picking up a bottle of Bio-Safe and a 1oz pack of Bio-Spira. Also, i will probably buy all of the 9 black phantom tetras, 1 bristlenose pleco and apple snail since i am able to add them all at once. thanks for the help.
