No, it doesn't matter how many you have. In fact, you don't need them at all provided your filters are providing sufficeint surface agitation. They can be used mainly if you like the look of them, as I do.

2. To agitate the water where I think I have low circulation. I find this out by grinding flake food finely and then releasing it into the flow of the water. If I notice that the food stays stagnant at one spot for a while, then I may have low agitation there. I had one spot like this before, so I bought an air stone to fix that problem.
