Just stick the tube end in the tank....run it out the door.... and give a good suck or two on the end that is outside...you can cup your hand around the end if you dont want to put your mouth on it.... you can also see the water coming since the tubing is clear so you know to get your mouth away before you get a mouthful
In my 55g I have to raise it over the center brace of the tank to get both sides when gravel vacuuming...if you have to do this just turn the valve on the tube end off....pull tube out and goto the other side of brace....dip tube end in so it fills with water and then turn valve back on and syphon should restart itself.

To get your python to start without it attached to the faucet is easy as pie. Extend the drain end of the hose to where you want it to go... in my case it's right out the window to the garden. Now put the vacuum end of the hose in the tank. Tip the end up so it fills with water. Lift the tube out of the tank so the water starts to drain out of the vacuum tube. AS SOON AS YOU SEE THE FLOW GOING BRING THE VAC TUBE BACK DOWN STILL TIPPED UP SO THE VAC FILLS... now under water you can tip the vac tube down and do your vacuuming. I have never had to suck the end of the hose using this method. It takes some getting used to and you have to be quick sometimes.

If you are not fast enough though, fill the suction tube as above, pinch the hose with the other hand about 2-3 feet down, lift tube up and let the water fill the hose to the pinch point. Refill the suction tube, then release the pinch point.
