my stand diy style...

I would double up the 2x4's, adding another 2x4 between the top and bottom frame would make a world of difference. If you have any electrical skills you could wire in some electrical boxes right into the stand. You could also wire up a set to a switch and use those for your pumps and heater. When it comes time to do water changes you just flip the switch and when your done flip it again.

I agree. I would also "sandwich" another 2x4 between the top and bottom.
Here is a pic of how I did mine if it helps, I made this for my 300.

If you skin it with 1/2 or thicker plywood you dont need to add diagonal braces. Plywood will provide the best lateral support. Use lots of wood glue and put a screw every 3" on the plywood and you'll be set.

Great! Well I'm going to double up those 2x4's, just to double check you mean the vertical peices, correct - I can see from the pic that you did both? Also I'm planning on doing at least 1/2 ply skin, but will cut holes for the droors and doors, will this compromise their given support?

I would double on both, just for overkill. If you cut holes for doors, it shouldnt decrease strength too much, but make the back as solid as possible. I left a gap at the top of mine to allow for wet/dry plumbing. I just used cheap plywood on the back and 1/2" hardwood plywood on the rest. Here is the back of it:
