I will be setting up a new tank (fish only live rock for now) this weekend. My tank is on the wall to the utility room, so I would like to keep all equipment behind the wall if possible. Is it possible to plumb the overflow box behind the wall? Meaning can the tube come from the tank, go through the wall and have the box behind the wall. Thanks.

Yep. Not a problem to setup, but you'll want to a) make sure you have covers for tubing to prevent snails and such from climbing in and slowing water movement between the main tank and the sump, and b) that you have enough head on the return. Straight lines are best for all plumbing lines--corners and slants slow water movement, and can allow bubbles to develop in the lines.
Head refers to the height the water has to be pushed from where the pump will sit to where it comes out. Basically, the higher the water has to go, the bigger the pump needs to be to retain good flow. Small pump+high head = slow water movement.
