if you are going to use Bio-Spira, you have to condition the water with either Marineland's Bio-Safe or Seachem's Prime. any other water conditioner may affect the way the bacteria consume the ammonia, as many water conditioners these days have added ingredients to help convert ammonia into a harmless state. that will interefere with the bacterias ability to comsume ammonia. Just follow the instructions on the package the way it says to. shake well, don't add into filter and don't add to one spot. just dump it in all around the tank. you can add almost your entire stock of fish right after, as the bacteria will be hungry for a source of ammonia. plecos should be added much later, as they don't do well in a newly established tank.
